Shares of Amylyx Pharmaceuticals experienced a significant rally of approximately 21% during Tuesday’s pre-market session following the company’s disclosure that avexitide, its experimental GLP-1 receptor antagonist, achieved success in a pivotal late-stage clinical study. The LUCIDITY Phase 3 study represented a 16-week investigation conducted across multiple centers using a randomized, double-blind, placebo-controlled design with 78 enrolled subjects. The research evaluated avexitide’s effectiveness in treating post-bariatric hypoglycemia, a serious metabolic complication characterized by dangerously low blood glucose levels following food consumption, typically emerging years after bariatric surgical procedures.
The study successfully achieved its primary objective, which had received prior regulatory alignment. Participants receiving avexitide experienced 55% fewer episodes of severe hypoglycemia compared to the placebo group.
Additionally, avexitide demonstrated positive outcomes across all secondary measures. These encompassed data gathered through both patient-administered fingerstick glucose tests and continuous glucose monitoring systems.
The mechanism of action for avexitide differs fundamentally from popular GLP-1 agonist medications such as Wegovy and Zepbound. Instead of mimicking GLP-1 hormone activity to reduce blood glucose, avexitide functions as an antagonist, blocking GLP-1 receptors located on pancreatic cells to prevent excessive hormonal signaling following meal intake.
Analyst Price Target Increase
Prior to the data release, Mizuho Securities elevated its price objective for AMLX from $24 to $30 while reaffirming its Outperform recommendation. The investment firm also designated the stock as a premier 2026 selection and projected a possible appreciation of $10 to $15 from existing levels contingent on favorable Phase 3 outcomes.
The FDA has granted avexitide both Breakthrough Therapy Designation and Orphan Drug status, regulatory classifications that may facilitate an accelerated approval process.
According to Amylyx, preliminary activities related to NDA submission readiness have commenced. The company is targeting a commercial debut in 2027.
Broader Market Backdrop
The positive movement occurred against challenging conditions in the wider equity markets. Pre-market indicators showed the Nasdaq declining 1.2% while the S&P 500 retreated 0.5%. Biotechnology stocks generally faced pressure during the session.
Company executives convened a conference call at 8:00 a.m. Eastern Time on Tuesday to review the trial outcomes with investors and analysts.
Amylyx had initially projected a broader Q3 2026 window for data disclosure. Tuesday’s release represented an acceleration of that timeline.
The pre-market valuation of $26.76 for AMLX shares represented a decisive break above the stock’s former 52-week high of $24.60.
