Shares of Merck & Co. experienced a modest retreat despite announcing encouraging Phase 3 clinical trial data for an innovative melanoma treatment created in partnership with Moderna. The stock dropped 0.67%, settling at $148.87 with a $1.01 decline as afternoon trading pressure offset morning momentum. The trial demonstrated meaningful reductions in both cancer recurrence rates and metastatic spread when participants received a personalized mRNA-based therapy combined with Merck’s established immunotherapy drug KEYTRUDA.
Clinical Trial Demonstrates Significant Reduction in Cancer Recurrence
The pharmaceutical giants evaluated intismeran autogene, an investigational therapy customized to match the specific genetic mutations present in individual patient tumors. This treatment leverages messenger RNA technology to train the immune system to identify and attack cancer-specific biomarkers. Scientists combined intismeran with Merck’s checkpoint inhibitor KEYTRUDA following surgical removal of melanoma tumors.
The INTerpath-001 study recruited 1,137 individuals diagnosed with fully resected melanoma ranging from stage IIB to stage IV. Trial participants received either the intismeran-KEYTRUDA combination or a placebo paired with KEYTRUDA. Following a median observation period of 24.6 months, data revealed the combination therapy decreased recurrence or mortality risk by 42%.
Additionally, the experimental regimen lowered the probability of distant metastatic spread or death by 38% compared to KEYTRUDA monotherapy. Both outcomes achieved statistical significance, with benefits appearing consistent across predetermined patient subgroup analyses. While overall survival statistics remain preliminary, early trends favor the combination approach.
Efficacy and Safety Profile Emerges After 24-Month Follow-Up
At the two-year mark, 82.2% of patients receiving the combination therapy remained recurrence-free, versus 70.0% of those treated with KEYTRUDA alone. For freedom from distant metastatic disease, the proportions stood at 88.5% and 80.2%, respectively. Neither cohort had yet reached the median timepoint for recurrence-free or distant metastasis-free survival.
Throughout the initial six-month period, investigators observed comparable recurrence and metastasis patterns between treatment arms. Yet a preplanned interim assessment at six months revealed a 58% reduction in recurrence or death risk favoring the combination. The same analysis documented a 50% decrease in distant metastatic spread or mortality risk.
Adverse events attributed to treatment emerged in 95.8% of combination recipients compared with 90.0% of the control arm. Severe grade 3–5 treatment-related complications occurred in 14.2% and 13.4% of patients, respectively. The most frequently reported reactions among combination therapy recipients included fever, pain at injection sites, and fatigue.
Partners Expand Development Program Across Multiple Cancer Types
Merck and Moderna intend to initiate regulatory discussions regarding these findings while continuing to gather long-term survival information. The collaborators will share comprehensive trial data at the 2026 European Society for Medical Oncology Congress. Publication of the complete results has been accepted by the New England Journal of Medicine.
The partnership currently operates nine Phase 2 and Phase 3 clinical investigations spanning multiple cancer indications and treatment settings. Three late-stage trials are evaluating the therapy in non-small cell lung cancer patients, while additional studies explore different clinical scenarios. Research teams are further investigating intismeran’s potential in pancreatic and gastric malignancies.
Previous Phase 2b melanoma data provided the foundation for advancing this personalized treatment approach into larger confirmatory trials. Despite the absence of mature overall survival data, the companies view these results as validation of their development strategy. Nevertheless, Merck’s stock price declined throughout the trading session despite the positive clinical developments and ongoing expansion of the program.
