Shares of ImmunityBio (IBRX) rose 1.12% to finish at $9.91 on Wednesday, climbing $0.11 amid fluctuating market conditions. The stock continued its upward momentum in extended trading, adding another 0.81% to reach $9.99 after the biotech firm revealed a significant regulatory milestone. The FDA has broadened ANKTIVA’s approved indication to encompass patients diagnosed with BCG-unresponsive non-muscle invasive bladder cancer (NMIBC) who present with papillary-only tumors.
Regulatory Agency Broadens ANKTIVA’s Patient Eligibility
The U.S. Food and Drug Administration has widened ANKTIVA’s approved use when administered alongside Bacillus Calmette-Guérin (BCG) therapy. This regulatory action incorporates adult patients with papillary-only tumors into the treatment’s eligible population. ImmunityBio can now provide this therapeutic combination to an expanded cohort of bladder cancer patients whose disease has become resistant to BCG treatment.
This recent regulatory decision builds upon the FDA’s original ANKTIVA authorization granted in April 2024. That initial clearance encompassed patients with BCG-unresponsive NMIBC featuring carcinoma in situ (CIS), whether accompanied by papillary tumors or not. Yet the original approval specifically left out patients presenting with papillary-only disease in the absence of CIS.
This broadened indication represents the first FDA clearance addressing this particular subset of bladder cancer patients. ImmunityBio now possesses regulatory approval for ANKTIVA combined with BCG across both CIS presentations and papillary-only manifestations. Moreover, this extended label substantially enlarges the therapy’s potential reach throughout the U.S. bladder cancer patient population.
Trial Evidence Underpins Broader Therapeutic Use
The FDA examined available scientific data demonstrating ANKTIVA’s efficacy in addressing BCG-unresponsive bladder cancer. This evidence base incorporated findings from Cohort A within ImmunityBio’s QUILT-3.032 clinical investigation. The study assessed ANKTIVA plus BCG in participants with CIS, whether papillary tumors were present or not.
Out of 77 enrolled patients, 62% demonstrated a complete response to the combination therapy during the clinical investigation. Additionally, 58% of those who responded sustained their response for a minimum of 12 months. Another 40% maintained therapeutic response for at least 24 months, illustrating durable treatment benefit in a significant portion of patients.
The therapy demonstrated a tolerable safety profile, with the majority of documented adverse events related to bladder symptoms. Frequently reported reactions encompassed painful urination, increased urinary frequency, and hematuria, typically at mild to moderate severity levels. Nevertheless, prescribing information cautions that postponing cystectomy surgery may permit potentially lethal metastatic disease progression.
Company Positions for Expanded Commercial Opportunity
Non-muscle invasive bladder cancer represents roughly 80% of all bladder cancer cases diagnosed across the United States. Moreover, between 85% and 90% of patients with newly diagnosed NMIBC exhibit papillary disease at presentation. These statistics underscore the considerable patient population impacted by this condition.
Individuals with high-grade papillary tumors encounter substantial risks once their cancer becomes refractory to BCG therapy. Disease recurrence and progression pose considerable challenges, especially for patients hoping to avoid radical cystectomy. Consequently, the expanded regulatory approval provides an additional therapeutic avenue for qualifying patients within this demographic.
ImmunityBio anticipates the wider indication will expand ANKTIVA’s addressable U.S. market beyond the original CIS patient subset. The biotechnology firm also seeks to accelerate commercial momentum following the therapy’s initial regulatory clearance in 2024. The latest FDA action reinforces ImmunityBio’s competitive positioning in bladder cancer immunotherapy while broadening its commercial prospects.
