BeOne Medicines unveiled clinical data showing a 36% response rate in advanced liver cancer patients during the ESMO Congress 2026 held in Madrid. The biotechnology firm disclosed updated findings spanning hepatocellular carcinoma, non-small cell lung cancer, and breast cancer programs. Despite these positive developments, BeOne Medicines AG (ONC) stock dropped $4.64, closing 1.21% lower at $377.63.
BGB-B2033 Delivers 36% Objective Response in Advanced Hepatocellular Carcinoma
BeOne Medicines shared refreshed Phase 1 data for BGB-B2033, a novel agent developed for patients with advanced hepatocellular carcinoma. The therapy produced a 36% confirmed objective response rate when administered at the dose selected for Phase 3 testing. This performance notably surpassed the roughly 5% response rates historically observed with specific late-stage tyrosine kinase inhibitors.
The trial enrolled 94 participants, of whom 93.6% had previously received PD-(L)1 immunotherapy treatment. An additional 61.7% had been exposed to both immunotherapy agents and tyrosine kinase inhibitors. Analysis at the recommended dose revealed an estimated 74.1% six-month confirmed response durability rate.
Tolerability data further bolstered the development program, with severe treatment-related adverse events occurring in just 5.3% of participants. Treatment discontinuation due to adverse reactions occurred in only 2.1% of patients across all evaluated dose cohorts. The company has indicated plans to initiate global registration-enabling trials before 2026 concludes.
Lung Cancer Portfolio Expands With PRMT5 Inhibitor and ADC Data
BeOne Medicines presented preliminary results for BGB-58067, an investigational PRMT5 inhibitor designed for MTAP-deficient malignancies. The compound generated a 25.3% confirmed response rate among 75 evaluable non-small cell lung cancer patients. Investigators also documented a 76% disease control rate, suggesting benefit beyond objective tumor shrinkage.
Among patients with EGFR wild-type disease, the treatment produced a 38.5% response rate. The overall non-small cell lung cancer population demonstrated a median progression-free survival of 7.7 months. Notably, patients harboring untreated, asymptomatic brain metastases showed a 30.8% intracranial response rate.
In parallel, BeOne disclosed positive early data for BG-C477, a proprietary antibody-drug conjugate targeting tumors expressing carcinoembryonic antigen. Among 25 patients with elevated CEA expression, the agent achieved a 56% confirmed response rate. The disease control rate reached 88%, supporting the company’s intention to advance development throughout 2027.
Global Phase 3 Breast Cancer Study Enrollment Underway for CDK4 Inhibitor
BeOne Medicines continues progressing BGB-43395, an investigational selective CDK4 inhibitor, through clinical development. The firm initiated patient enrollment in the international Phase 3 KANDELA-302 trial during May 2026. This study recruits patients diagnosed with HR-positive, HER2-negative advanced or metastatic breast cancer in the first-line setting.
The trial design positions BGB-43395 combined with letrozole against three approved CDK4/6 inhibitors paired with letrozole. The comparator regimens feature abemaciclib, palbociclib, and ribociclib, each representing standard-of-care options in this patient population. Investigators designated progression-free survival as the study’s primary outcome measure and overall survival as a critical secondary endpoint.
BeOne continues building its solid tumor franchise alongside its existing hematologic malignancy business. The FDA has previously conferred Fast Track and Orphan Drug status upon BGB-B2033 for hepatocellular carcinoma. The collective clinical updates represent meaningful progress across multiple programs, though confirmatory data from larger, controlled trials remains necessary.
